Orphelin de père à l’âge de 12 ans, il exerce diverses activités : apprenti typographe, rédacteur d’articles dans le journal de son frère, pilote de bateau à vapeur sur le Mississippi. C'est de cette époque que vient son pseudonyme : alors qu'il ramène la corde de sondage pour vérifier la profondeur du fleuve, son capitaine lui crie : "Mark Twain !, Mark Twain !", c'est-à-dire : "Marque deux sondes ("profondeur suffisante", en anglais "safe water").
